    Protein Summary

    The gene 15608974 from M. tuberculosis encodes the malate synthase PF01274 EC:2.3.3.9 COG2225 (alternative names: glyoxylate transacetase, malate synthetase, malic synthetase).  The structure of the enzyme from the same organism has been previously solved 1N8I.  The enzyme belongs to the class of alpha and beta (a+b) proteins and reveals TIM beta/alpha-barrel fold type SCOP51350.  This enzyme belongs to the family of transferases, specifically those acyltransferases that convert acyl groups into alkyl groups on transfer.   The enzyme catalyzes the reaction: cetyl-CoA + H2O + glyoxylate --> (S)-malate + CoA.  The enzyme participates in pyruvate metabolism and glyoxylate and dicarboxylate metabolism.  This is part of a bypass of the tricarboxylic acid cycle in which isocitrate lyase and malate synthase (GlcB) catalyze the net incorporation of carbon during growth of microorganisms on acetate or fatty acids as the primary carbon source.  Establishment or maintenance of a persistent infection by Mycobacterium tuberculosis requires the glyoxylate pathway [Ref].
