    Protein Summary

    Pfam Update: 

    This matches our DUF1735 at 67-127. Current we do not have any other surrounding domains for this sequence. This family does not have any know structures currently. We have no functional information, but it often found with glycohydrolases, so it _may_ be a carbohydrate binding domain. Will be interesting to follow this family up.
    Update: Following up what Rob has already detailed, there are a couple of structures within the family that I have now built from this, DUF4361 PF14274, ie 3NQK and 3P02. DALI searches of these throw up a weak match to 3poh which is a longer glycosyl-hydrolase_18 family member, and parts of the structures do superimpose onto each other. Otherwise all members are uncharacterised proteins.

The structure of target ID 416718 represents the first structure determined for this Pfam DUF1735 class.

    The structure of target ID 416718 was determined at a resolution of 2.61 Ang by 3 wavelength MAD phasing from a P321 crystal form with one subunit in the crystallographic asymmetric unit.

    Shown above is a ribbons representation of the structure of the monomer of target ID 416718. The ribbons are color-coded with the N-terminal end in blue and the C-terminal end in red.Each monomer consists of two domains. There us an N-terminal domain (residues 31-179) and a C-terminal domain (residues 180-339). The structure of the N-terminal domains shows structural similarity to those proteins belonging to the Clathrin adaptor appendage domain SCOP superfamily.

     

    Analytical size exclusion chromatography with static light scattering (SEC/SLS) supports the assignment of a trimer as a significant oligomerization state. The trimer is most likely represented by application of crystallographic three-fold symmetry to the monomer. A ribbons representation of the trimer is shown below with each monomer in a different color.
