    Protein Summary

    Gene phzB2 from Pseudomonas aeruginosa PAO1 encodes the NP_250591 protein that belongs to the phenazine biosynthesis protein A/B group (PF03284). Analysis of its genome context indicates a functional link with the phenazine biosynthesis proteins PhzC, D and E (score 0.77).

    PhzB shares similarity with both subunits of anthranilate synthase and the phzB open reading frame complemented an E. coli trpE mutant deficient in anthranilate synthase activity.[Ref] Products of the phzA and phzB genes are highly homologous to each other and may be involved in stabilization of a putative PCA-synthesizing multienzyme complex.[Ref] The results of cluster analyses indicated that the products of the phzA and phzB genes of P. aeruginosa strain PA14 most closely resemble PhzA2 and PhzB2 from P. aeruginosa PAO1.[Ref]

    Pre-SCOP classifies 3ff0 in the alpha+beta class, NTF2-like superfamily, hypothetical protein YesE family. SSM and DALI search hits are with the phenazine biosynthesis related proteins 3jum, 3dzl and 3b4o (Z=25), the NTF-2 like protein of unknown function 3ebt (1.9A rmsd, 118 Ca aligned, 11% id; Z=13) and 3ec9 (2.15A rmsd, 118 Ca aligned, 12% id; Z=13), as well as with a putative isomerase YesE 1s5a (2.12A rmsd, 123 Ca aligned, 21%id; Z=16).

    3ff0 assembles as a dimer in the crystallographic asymmetric unit, which is also its biological relevant state. Interestingly, an unknown ligans (UNL) is found in the putative active site of each protomers. The density is similar in shape to nitrobenzene or benzoic acid.
